Resumen ANTECEDENTES: El angiomixoma agresivo profundo perineal es un tumor mesenquimatoso de muy limitada aparición que se origina debido a un crecimiento tumoral del tejido conjuntivo que se expande a pesar de su naturaleza benigna; se caracteriza por un comportamiento agresivo. CASO CLÍNICO: Paciente de 38 años, con un nódulo genital de 3 cm en el labio mayor izquierdo, con sospecha clínica de quiste de la glándula de Bartolino. Durante la intervención para su drenaje se objetivó una tumoración blanda, de aspecto mesenquimal, pediculado. El análisis histológico confirmó que se trataba de un angiomixoma agresivo profundo perineal. Posterior al estudio de extensión negativo, se completó la vulvectomía simple izquierda, con un posoperatorio favorable. CONCLUSIONES: La obtención de una biopsia inicial es decisiva, sobre todo en casos de tumores mesenquimales agresivos, como el angiomixoma agresivo perineal profundo. De esta manera puede establecerse un plan de tratamiento individual en función del diagnóstico histológico definitivo.


Abstract BACKGROUND: Aggressive deep perineal angiomyxoma is a mesenchymal tumor of very limited occurrence that originates due to a tumorous growth of connective tissue that expands despite its benign nature; it is characterized by an aggressive behavior. CLINICAL CASE: 38-year-old patient with a 3 cm genital nodule on the left labium majus, with clinical suspicion of Bartholin's gland cyst. During the intervention for its drainage, a soft, mesenchymal, pedunculated tumor was observed. Histological analysis confirmed that it was an aggressive deep perineal angiomyxoma. Following a negative extension study, a simple left vulvectomy was completed, with a favorable postoperative course. CONCLUSIONS: Obtaining an initial biopsy is critical, especially in cases of aggressive mesenchymal tumors, such as deep perineal aggressive angiomyxoma. In this way an individual treatment plan can be established based on the definitive histologic diagnosis.

ABSTRACT

ABSTRACT The tarwi is an Andean legume with a high nutritional value from which a vegetable beverage can be obtained. Cereals, like oat, have good characteristics as a prebiotic for the production of functional drinks, whose consumption is currently increasing. The objective of the research was to design a probiotic fermented beverage based on fresh milk, tarwi beverage, and oatmeal. An optimal treatment consisted of 1.9% oatmeal, 39.9% tarwi beverage, 46.2% fresh milk, 10.0% honey, and 2.0% probiotic culture; determined by applying a rotatable central composite design of surface response methodology. It had a probiotic count of 3.47x108 cfu/mL, a protein content of 3.75%, and overall acceptability of 7 points, which corresponds to "I like it very much". The result was experimentally validated. Likewise, the shelf life of the optimal beverage was 20 days at 5 °C with appropriate functional, nutritional, and sensory characteristics.


RESUMEN El tarwi es una leguminosa andina con un alto valor nutricional a partir de la cual se puede obtener una bebida vegetal, los cereales como la avena tienen mejores características como prebióticos para la producción de bebidas funcionales, cuyo consumo está aumentando actualmente. El objetivo de la investigación fue diseñar la formulación de una bebida fermentada probiótica a base de leche fresca, bebida de tarwi y avena. Se determinó un tratamiento óptimo que consistió en 1,9% de avena, 39,9% de bebida de tarwi, 46,2% de leche fresca, 10,0% de miel y 2,0% de cultivo probiótico; mediante la aplicación de un diseño compuesto central rotable de metodología de respuesta de superficie. Se reportó un recuento de probióticos de 3,47x108 ufc/mL, un contenido de proteínas de 3,8% y una aceptabilidad general de 7 puntos, que corresponde a "Me gusta mucho"; el resultado fue validado experimentalmente. Asimismo, la vida útil de la bebida óptima fue de 20 días de almacenamiento a 5 °C con características funcionales, nutricionales y sensoriales apropiadas.

Background: Asparagus (Asparagus officinalis L.) green is a vegetable with a great demand worldwide, and likewise, it is highly perishable, due to its high respiration rate that accelerates its senescence. Disinfection of vegetables after their harvest is an obligatory practice that can reduce losses by decomposition due to the attack of microorganisms. Therefore, it is vital to preserving its microbiological and sensory characteristics to reach the final consumer. Objective: to evaluate the effect of gaseous ozone (0 to 10 ppm) and storage time (0 to 30 days) on phenol content, overall appearance, count of molds, psychrophilic bacteria, and viable mesophilic aerobes. Methods: the response surface methodology was used, applying a rotatable central composite design. Results: the results indicated that there was a significant influence (p <0.05) of the independent variables on the characteristics studied, as well as an adequate lack of fit of the quadratic regression model (p> 0.05). By means of the contour superposition technique, it was determined that the optimal conditions for the highest retention of phenol content (16.99 mg/g) and overall appearance (7.61 points) and lower counts of viable aerobic mesophilic bacteria (5.3 x 103 CFU/g) they corresponded to 10 ppm of gaseous ozone up to 25.91 days of storage, with adequate quality characteristics in the spears. Conclusion: the region of interest was determined for optimal retention of phenol content and overall appearance, and a lower count of viable aerobic mesophilic bacteria in green asparagus during postharvest, suggesting to use the initial application of ozone gas at 10 ppm allowing 25.9 days storage at 1 °C. The results indicate that this technology is a good alternative in the conservation of fresh vegetables


Antecedentes: El espárrago (Asparagus officinalis L.) verde; es una hortaliza con una gran demanda a nivel mundial, y, asimismo, es altamente perecible, por su elevada velocidad de respiración que, acelera su proceso de senescencia. La desinfección de los vegetales después de su cosecha es una práctica obligada que puede disminuir las pérdidas por descomposición debido al ataque de microrganismos. Por lo tanto, es muy importante conservar sus características microbiológicas y sensoriales para llegar al consumidor final. Objetivo: evaluar el efecto del ozono gaseoso (0 a 10 ppm) y tiempo de almacenamiento (0 a 30 días) sobre el contenido de fenoles, apariencia general, recuento de mohos, bacterias psicrófilas y aerobias mesófilas viables. Métodos: se utilizó la metodología de superficie de respuesta, aplicando un diseño compuesto central rotable. Resultados: los resultados indicaron que existió influencia significativa (p<0.05) de las variables independientes sobre las características estudiadas, así como, una adecuada bondad de ajuste del modelo de regresión cuadrático (p>0.05). Mediante la técnica de superposición de contornos se determinó que las condiciones óptimas para la mayor retención de contenido de fenoles (16.99 mg/g) y apariencia general (7.61 puntos) y menor recuentos de bacterias aerobias mesófilas viables (5.3 x 103 UFC/g) correspondieron a 10 ppm de ozono gaseoso hasta los 25.91 días de almacenamiento, con adecuadas características de calidad en los turiones. Conclusión: se determinó la región de interés para una óptima retención de contenido de fenoles y apariencia general, así como, menor recuento de bacterias aerobias mesófilas viables en el espárrago verde durante la postcosecha, sugiriendo utilizar la aplicación inicial de ozono gaseoso a 10 ppm permitiendo 25.9 días de almacenamiento a 1 °C. Los resultados indican que esta tecnología es una buena alternativa en la conservación de hortalizas frescas

ABSTRACT

Identificar el sesgo de confusión y cómo controlarlo es uno de los desafíos metodológicos más importantes en el diseño de estudios que buscan identificar la causalidad. Este sesgo está presente en cualquier análisis de la asociación entre una exposición y un resultado de interés, una asociación que puede estar sesgada o no por una tercera variable llamada confusor. Podemos diagnosticar un confusor en todos los casos en los que este crea una asociación espuria entre una variable de exposición o variable independiente y la variable de resultado o variable dependiente. Para controlar el sesgo de confusión, podemos usar diferentes métodos. Estos incluyen aquellas técnicas aplicadas en el diseño del estudio, tales como restricción, aleatorización y coincidencia, y aquellas técnicas empleadas en el análisis de datos, como la estratificación, el análisis multivariado, la estandarización, los puntajes de propensión, el análisis de sensibilidad y el inverso ponderación de probabilidad. En esta revisión, analizamos cómo identificar una variable de confusión y las principales técnicas para controlar el sesgo de confusión.


Addressing confounding bias is one of the challenges when conducting causality studies. This occurs when we report a causal association between an exposure and an outcome, when in fact it could be result of the effect of a third factor called confounding variable. That is, when a confounder variable creates a spurious relationship between the exposure or independent variable and the outcome of interest or dependent variable. By knowing the confounding variables and their association with the exposure of interest, the confounding bias could be controlled. To control for confounding bias, we can use different methods. These include techniques applied in study design, such as restriction, randomization, and coincidence, and techniques used in data analysis, such as stratification, multivariate analysis, standardization, propensity scores, analysis sensitivity and the inverse probability weighting. In this review, we discuss how to identify a confounding variable and the main techniques for controlling for confounding bias.

ABSTRACT

O objetivo deste estudo foi investigar o papel do fator de crescimento derivado de plaquetas-BB (PDGF-BB) na concentração de 300ng/ml na taxa de proliferação e adesão de células derivadas da granulação óssea humana a fragmentos radiculares periodontalmente comprometidos. Na primeira etapa do estudo, foi estabelecida cultura primária de células da granulação óssea de dois pacientes adultos, sistemicamente saudáveis, não fumantes. Após a expansão celular, as células foram caracterizadas para determinação do fenótipo por meio de ensaios de viabilidade celular, MTT, ensaio de atividade de fosfatase alcalina, ensaio de mineralização e caracterização imunohistoquímica por meio de citometria de fluxo (segunda etapa). Na terceira etapa do estudo, os efeitos da adição de PDGF-BB recombinante humano na concentração de 300ng/ml na taxa de proliferação e adesão de células derivadas da granulação óssea a superfícies radiculares periodontalmente comprometidas foram investigados. A taxa de proliferação celular estimulada pelo PDGF-BB (grupo teste) ou pelo meio de cultura (grupo controle) foi investigada por meio de contagem de células viáveis nos frascos de cultura após 1, 3, 5 e 7 dias do cultivo celular. Foram obtidos 30 fragmentos dentários a partir de dentes extraídos por razões periodontais. Os fragmentos foram raspados com curetas Gracey e condicionados com solução em gel de EDTA a 24% durante 3 minutos, lavados com solução de soro fisiológico, secos e posicionados em placas de 24 poços. Foram incubadas sobre os fragmentos tratados 1x104 células GO por 24 horas, seguido por fixação e preparo para análise por microscopia eletrônica de varredura (MEV). O número de células aderidas sobre os fragmentos foi analisado nas fotomicrografias. O padrão de crescimento das células GO foi compatível com células ósseas, com modificação do padrão do crescimento com o aumento do número de passagens. Houve atividade de fosfatase alcalina em meio osteogênico e convencional, com pico máximo aos 7 dias e atividade de mineralização estimulada ou não por meio osteogênico, com pico máximo aos 21 dias. A análise por meio de citometria de fluxo demonstrou que as células GO não expressaram CD105 e CD166 na 14a passagem, indicando sua diferenciação celular avançada nesse período. A adição de rhPDGF-BB resultou em mudança na taxa de proliferação celular, observando-se pico máximo de crescimento aos 7 dias, com diferenças estatisticamente significantes (p < 0.005; ANOVA post hoc Tukey) em relação aos períodos de 1, 3 e 5 dias. O ensaio de MTT demonstrou maior viabilidade celular no período de 48 hs, comparativamente aos períodos de 24 e 72 horas, quando a densidade óptica celular diminuiu de forma significativa (p< 0.05; Friedmann pósteste Dunn). No ensaio de adesão celular, pode-se observar que a adição de rhPDGFBB aumentou significativamente o número de células aderidas aos fragmentos dentários (p< 0.05; teste t não pareado com correção Welch), com alteração da morfologia celular. Esses resultados sugerem que as células GO tem características compatíveis com linhagem de células osteoblásticas, de fenótipo mais diferenciado após a 12a passagem. A adição de rhPDGF-BB (300ng/ml) resulta em aumento da taxa de proliferação das células GO e do número de células aderidas a fragmentos radiculares, indicando que, nesta concentração, o fator de crescimento é citocompatível, favorecendo a proliferação e adesão celular.(AU)


The goal of this study was to investigate the effects of recombinant human platelet derived growth factor (rhPDGF-BB) at the concentration of 300ng/ml in the proliferation and adhesion of human bone granulation cells to periodontally diseased root fragments. At the first stage of the study, the granulation tissue existent in healing sockets (21 days after its creation) was collected from two systemically healthy nonsmoking adults to the establishment of primary culture. The in vitro properties of bone granulation (BG) cell lineage were characterized by cell viability, MTT, alkaline phosphatase activity and mineralization assays. The effects of culture medium (control) and rhPGDF-BB 300ng/ml (test) in the proliferation and adhesion of BG cells were investigated. The rate of BG cells proliferation was investigated by the number of viable cells present at 1, 3, 5 and 7 days after platting. Thirty root fragments were obtained from teeth extracted for periodontal reasons. Root fragments were scaled and root planed, conditioned with EDTA 24% for 3 minutes, rinsed in saline solution, air-dryed and positioned in 24-well plates. Each fragment was seeded with 104 BG cells, fixated after 24 hours and prepared for analysis in SEM. The number of cells adhered to the fragments was analysed in photomicrographies. BG cells growth pattern was compatible with osteogenic cell lineage, showing modification with the increasing number of cell passage. GO cells expressed alkaline phosphatase activity in conventional and osteogenic culture medium, with maximum peak at 7 days, as well as mineralization activity stimulated or not by osteogenic or non-osteogenic culture medium, with maximum peak at 21 days. The analysis by flow cytometer showed that BG cells have not expressed CD105 and CD106 at the 14th passage, indicating its advanced cell differentiation. The addition of rhPDGF-BB resulted in modification of proliferation rate, with maximum peak observed at 7 days, significantly different from 1-, 3- and 5-day periods (p< 0.005; ANOVA post hoc Tukey). MTT assay showed greater cell viability after 48 hours than after 24 and 72 hours, when optical density has significantly diminished (p< 0.05; Friedmann post hoc Dunn). At cell adhesion assay, it could be observed that the adhesion of rhPDGF-BB has significantly increased the number of cells adhered to root fragments (p< 0.05; unpaired t test with Welchs correction), and alterations in cell morphology. These results suggest that BG cells present in vitro characteristics compatible with osteoblastic cell lineages, with a more differentiated phenotype after the 12th passage. The addition of rhPDGF-BB (300 ng/ml) results in increase of the rate of BG cell proliferation and in the number of cells adhered to root fragments, indicating that, at this concentration, the growth factor is compatible with BG cells and favors cells proliferation and adhesion.(AU)

During menopause, changes occur in the laryngeal structures that have implications for the voice. The purpose of this study was to determine the effect of induced menopause on the morphological parameters of the vocal fold mucosa in rats. Ten adult female Sprague-Dawley rats were used as samples and divided into two groups: 5 were surgically oophorectomized (OVX), and 5 underwent a surgical procedure similar to oophorectomy but without removal of the ovaries (SHAM). After 30 days surgery, the characteristics of the epithelium that forms the vocal fold mucosa in terms of cellular arrangement and organization of the epithelium were observed. Through the Morphometrics XS software, the epithelial height and the number and density of cellular layers were determined. Our results indicate that there were alterations in the number of cell layers that constitute the epithelium, as well as features, such as cellular cohesion and increased extracellular matrix. The number of cell layers was significantly higher (p < 0.01) in the SHAM group with 6.66 (SD 1.07), whereas in the OVX was 3.2 (SD 0.83). The average thickness of the epithelium was 534.71 mm (SD 119.89), whereas in the SHAM group was 486.84 mm (SD 82.95); these differences were not statistically significant (p = 0.112). Changes in the characteristics of the epithelium covering the vocal folds can be related to clinical abnormalities, such as reduced voice quality and degeneration of the vocal folds in postmenopausal women.


Durante la menopausia ocurren cambios en las estructuras laríngeas que tienen implicancias en la voz. El objetivo de este estudio es determinar el efecto de la menopausia inducida sobre parámetros morfológicos de la mucosa de los pliegues vocales en ratas de laboratorio. Se analizaron 10 ratas Sprague-Dowley hembras adultas, divididas en dos grupos, 5 fueron ooforectomizadas quirúrgicamente (OVX) y 5 fueron sometidas a un procedimiento SHAM (SHAM). Luego de 30 días postcirugía, se observaron las características del epitelio del pliegue vocal, en términos de disposición celular y organización del epitelio. Adicionalmente con la ayuda del software Morphometrics XS se determinó la altura del epitelio y la densidad de número de estratos celulares. Nuestros resultados indican que existen alteraciones en número de estratos celulares, así como también en características como cohesión celular, acompañado de un aumento de la matriz extracelular. El número de estratos celulares fue significativamente mayor (p<0,01) en el grupo SHAM con 6,66 (DS 1,07), mientras que en el OVX fue de 3,2 (DS 0,83). El grosor medio del epitelio fue de 534,71 µm (DS 119,89), mientras que en el grupo SHAM fue de 486,84 µm (DS 82,95), estas diferencias no fueron estadísticamente significativas (p=0,112). Se sugiere que alteraciones en las características del epitelio que reviste los pliegues vocales pueden relacionarse con alteraciones clínicas como la disminución de la calidad vocal y la presbifonía características de la mujer postmenopáusica.
